MOTS-c Dosage Calculator

Free MOTS-C reconstitution calculator. Enter vial size (5-10 mg), BAC water volume, and dose to get concentration, syringe units, and doses per vial. Includes bolus, intermittent, and low-dose daily protocols plus exercise-timing guidance. Verify MOTS-C quality with independent lab test data on Peptigrity.

A 10 mg vial of MOTS-c reconstituted with 2 mL of bacteriostatic water gives a concentration of 5.0 mg/mL, or 50 mcg per unit on a U-100 syringe. For a 5 mg dose, draw the syringe to 100.0 units (1.00 mL). At that dose the vial provides about 2 doses.

Starts at a 10 mg vial and 5 mg; 5 mg is the amount this page's own reference tables use. The calculator picks the water so your dose lands on a line of your syringe — or set your own. Arithmetic only, not a recommendation.

Why 2 mL? It’s the amount this calculator starts with, and each 5 mg dose already lands on the 100 line of your 1 mL syringe — a numbered line.
How we pick the water
  1. It fits your vial, and it’s at least 1 mL.
  2. One dose fits one syringe and is at least 5 units.
  3. The dose lands on a line of your syringe.
  4. If the amount this calculator starts with already does that, we keep it.
  5. Otherwise: whole mL first, then half, then 0.1 mL steps.
  6. A numbered line if possible, 10 to 50 units if possible.
  7. Easy math: 10 units come out as a round number.
  8. Less water, if it’s still a tie.
We only pick the water. The dose is always yours.

MOTS-C is a mitochondrial-derived peptide (MDP) - a 16-amino-acid signalling molecule encoded within the 12S rRNA gene of mitochondrial DNA. Its primary mechanism is AMPK activation, making it one of the few peptides characterised as an “exercise mimetic.” Doses per administration are high relative to other research peptides.

Protocol 1 - Bolus (Weekly)

ParameterValue
Dose5-10 mg per injection
Frequency1× weekly
Duration8-12 weeks
TimingMorning, 30-60 min before exercise
NotesMost closely mirrors preclinical dosing; large draw volume

Protocol 2 - Intermittent (2-3× weekly)

ParameterValue
Dose5 mg per injection
Frequency2-3× weekly (e.g., Mon/Wed/Fri)
Duration6-8 weeks
TimingMorning, before activity
NotesHigher weekly total; used by those seeking more consistent AMPK activation

Protocol 3 - Low-Dose Daily

ParameterValue
Dose1-2 mg daily
Frequency1× daily
Duration8-12 weeks
TimingMorning
NotesHighest total consumption; anecdotally gentler onset

MOTS-C Reconstitution - Quick Reference

MOTS-C has high per-dose requirements - a single 5 mg dose uses half of a 10 mg vial. Choose BAC water volume carefully to keep draw volumes manageable.

VialBAC WaterConc.5 mg Draw10 mg DrawDoses (5 mg)
10 mg1 mL10 mg/mL50 units100 units (full)2

5 mg0.5 mL10 mg/mL50 unitsN/A1
5 mg1 mL5 mg/mL100 units (full)N/A1

Common: 10 mg vial + 1 mL BAC water = 10 mg/mL. A 5 mg dose = 50 units (half a syringe). Each 10 mg vial = exactly 2 bolus doses. For reconstitution technique, see reconstituting peptides step by step.

Timing and Exercise

TimingRationaleCommunity Usage
30-60 min before exerciseAMPK activation may synergise with exercise-induced AMPKMost common
Morning, non-exercise daysMaintains consistent administration scheduleCommon for daily protocols
Evening-Uncommon - some report energy increase that may affect sleep

MOTS-C and Weight Management

MOTS-C activates AMPK - the same pathway triggered by exercise and metformin - which regulates glucose uptake, fatty acid oxidation, and mitochondrial biogenesis. Preclinical studies in mice showed MOTS-C prevented high-fat-diet-induced obesity and improved insulin sensitivity. Human data is extremely limited; plasma MOTS-C levels have been correlated inversely with BMI in observational studies, but no controlled human weight loss trials have been published.

WADA Prohibited Status

MOTS-C is classified under S2 - Peptide Hormones, Growth Factors, Related Substances and Mimetics on the WADA Prohibited List. It is banned both in-competition and out-of-competition. Athletes subject to anti-doping regulations should not use MOTS-C. Detection methods based on mass spectrometry can identify MOTS-C in urine and blood samples.

How to Verify MOTS-C Quality

MOTS-C is a 16-amino-acid peptide with a molecular weight of approximately 2,174.7 Da. Check the CoA for ≥98% HPLC purity and mass spectrometry confirming the expected molecular weight. Cross-reference with independent lab test results. For CoA interpretation, see red flags in peptide certificates of analysis.

Frequently asked questions

How many units is 5 mg of MOTS-C?

At 10 mg/mL (10 mg vial + 1 mL BAC water): 5 mg = 50 units. At 5 mg/mL (10 mg + 2 mL): 5 mg = 100 units (a full syringe). Use 1 mL BAC water for manageable draws.

Should I take MOTS-C daily or weekly?

Three protocols exist: weekly bolus (5-10 mg once), intermittent (5 mg 2-3× weekly), and low-dose daily (1-2 mg). The bolus protocol most closely mirrors preclinical dosing.

Is MOTS-C banned in sports?

Yes. MOTS-C is on the WADA Prohibited List (S2 - Peptide Hormones). Prohibited in-competition and out-of-competition. Athletes subject to testing should not use MOTS-C.

When should I take MOTS-C?

Morning, 30-60 minutes before exercise is the most common timing. MOTS-C activates AMPK, which may synergise with exercise. Some users report increased energy that could interfere with sleep if taken in the evening.

How many vials do I need for a full cycle?

Bolus (5 mg weekly, 8 weeks) = 4 × 10 mg vials. Intermittent (5 mg 3×/week, 6 weeks) = 9 vials. Daily (2 mg, 8 weeks) = ~12 vials. MOTS-C is one of the highest-consumption peptides by total milligrams.

Does MOTS-C help with weight loss?

Preclinical data shows MOTS-C prevented obesity in high-fat-diet mouse models and improved insulin sensitivity via AMPK activation. Human weight loss trials have not been published. It is not comparable to FDA-approved GLP-1 agonists like semaglutide.
