The Mitochondrial Blend combines three research compounds that each touch cellular energy metabolism: 5-Amino-1MQ (an NNMT inhibitor studied for metabolic/fat effects), MOTS-c (a mitochondrial-derived peptide involved in metabolic regulation and insulin sensitivity), and NAD⁺ (the central cofactor for cellular energy and sirtuin activity). Why are these three combined?
The rationale is convergent: NAD⁺ replenishes the coenzyme that mitochondrial energy production and sirtuins depend on; in preclinical (mouse) studies MOTS-c improved metabolic homeostasis and insulin sensitivity; and 5-Amino-1MQ inhibits NNMT, an enzyme whose activity is linked to adipose expansion and lowered cellular NAD⁺/methylation capacity — so in principle the three reinforce a common "cellular energy / metabolic" theme. These are research directions, not proven combined outcomes.
What's the evidence and status?
Each component has its own (mostly preclinical or early) research base, but the combination has not been validated in controlled human trials as of August 2026 — it is an experiential/research stack, not an approved therapy. What is the regulatory status of the Mitochondrial Blend's components?
None of the three components is approved by the FDA for human use as of August 2026. MOTS-c was among the peptides the FDA removed from its 503A Category 2 bulk-substances list in April 2026 after the nomination was withdrawn by the nominator — a removal that did not add it to Category 1 or authorise compounding. At its July 23–24, 2026 meeting the FDA's Pharmacy Compounding Advisory Committee voted to recommend MOTS-c (7–5–2) for the 503A compounding list — an advisory recommendation FDA reviewers opposed and the agency has not finalized; formal rulemaking is not expected before 2027. NAD⁺ is sold as a dietary-supplement ingredient rather than an approved drug, and 5-Amino-1MQ is a research-use-only compound with no regulatory approval anywhere. In sport, the two unapproved compounds (MOTS-c, 5-Amino-1MQ) fall under WADA's catch-all S0 category; NAD⁺ is not itself WADA-prohibited. Evidence level: regulatory status only — none FDA-approved; MOTS-c preclinical, NAD⁺ precursor RCTs show no clear functional benefit, 5-Amino-1MQ preclinical; the blend has no controlled human trial.
